Getting started

VENCLEXTA is an oral medicine taken by mouth each day. VENCLEXTA is taken in combination with azacitidine, or decitabine, or low-dose cytarabine. Your healthcare provider can advise which VENCLEXTA combination regimen may be appropriate for you.

You will start taking VENCLEXTA at a low dose and gradually increase to a higher dose—this is called a “ramp-up period.” The ramp-up dosing will depend on which regimen you and your healthcare provider have chosen. Once you’re at the full dose, continue to take VENCLEXTA as prescribed by your healthcare provider.

VENCLEXTA oral dosing schedule

Day 1

 

Take 100 mg of VENCLEXTA

and your healthcare provider will start you on azacitidine 75 mg/m2 (IV or SC), or decitabine 20 mg/m2 IV), or cytarabine 20 mg/m2 (SC) as determined by your healthcare provider.

Day 2

Take 200 mg of VENCLEXTA

in combination with azacitidine 75 mg/m2 (IV or SC), or decitabine 20 mg/m2 (IV), or cytarabine 20 mg/m2 (SC) as determined by your healthcare provider.

Day 3

Take 400 mg of VENCLEXTA

in combination with azacitidine 75 mg/m2 (IV or SC), or decitabine 20 mg/m2 (IV), or cytarabine 20 mg/m2 (SC) as determined by your healthcare provider.

Day 4 and beyond (28-day cycle)

In combination with azacitidine or decitabine:

Take 400 mg of VENCLEXTA

in combination with azacitidine 75 mg/m2 (IV or SC) on Days 1–7, or decitabine 20 mg/m2 (IV) on Days 1–5 of each 28-day cycle as determined by your healthcare provider.

OR

In combination with low-dose cytarabine:

Take 600 mg of VENCLEXTA

in combination with cytarabine 20 mg/m2 (SC) on Days 1–10 of each
28-day cycle as determined by your healthcare provider.

Your dosing schedule will be determined by your healthcare provider. Dosing schedules may differ from the examples above if you take medications that could interact with VENCLEXTA.

The recommended dose of VENCLEXTA may be delivered using any of the approved tablet strengths (eg, patients can take 2 x 50 mg tablets or 10 x 10 mg tablets instead of 1 x 100 mg tablet as needed).

Intravenous (IV) means within a vein. In this type of injection, the medication is injected directly into the vein.

Subcutaneous (SC) means under the skin. In this type of injection, the medication is injected into the tissue layer between the skin and the muscle.

Why is bone marrow biopsy important?

A test called a bone marrow biopsy that you may have done will help your healthcare provider monitor your progress. The results of this important test will help your provider determine if modifying your treatment is appropriate.

After your biopsy evaluation, your healthcare provider may require you to take a short treatment break or decide you can continue to the next cycle.

Bone marrow biopsy is a procedure in which a small sample of bone with bone marrow inside it is removed, usually from the hip bone.
